What is ergothioneine

2-mercaptohistidine trimethyl-betaine · unique sulfur amino acid · synthesized only by mushrooms, bacteria and some algae · NOT by mammals.

Ergothioneine (ERG) is a tetracyclic sulfur amino acid (histidine-derived) synthesized exclusively by mushrooms, mycobacteria and cyanobacteria. Humans cannot produce it · we obtain it only from diet and microbiota. Its unique characteristic: we have a specific transporter (OCTN1, SLC22A4 gene) that actively concentrates it in tissues of high turnover and oxidative stress (liver, kidney, retina, brain, bone marrow, erythrocytes).

That a nutrient has a dedicated transporter in humans strongly suggests biological function. Bruce Ames and others have proposed it as «longevity vitamin» (proposed but not classified vitamin). It drops with age. Low levels are associated with cognitive decline (Cheah-Halliwell, Singapore 2020), frailty and mortality. The richest food source is king oyster (Pleurotus eryngii) with ~5 mg/100 g · exceeds white button mushroom (Agaricus) ~1 mg/100 g.

Do I need to supplement if I eat mushrooms?
Probably not if you eat >3 servings/week varied mushrooms (button, portobello, shiitake, king oyster). One 100 g king oyster serving ~5 mg ERG = canon dose. Supplement if restricted diet or aggressive cognition/longevity protocol.
Is it the same as medicinal mushrooms (reishi, chaga)?
No. Medicinal mushrooms (reishi, chaga, cordyceps) provide other compounds (triterpenes, beta-glucans, polysaccharides). Only Pleurotus eryngii (king oyster) and some Hericium have ERG in high quantity. For specific ERG: king oyster or dedicated supplement.
Why do they call it "longevity vitamin"?
Bruce Ames + Barry Halliwell proposed it because it meets 4 criteria: 1. We don't synthesize it. 2. We have dedicated transporter (OCTN1). 3. Concentrates in high oxidative stress tissues. 4.
