The endocannabinoid system (ECS) is a body-wide lipid signaling network that regulates homeostasis: the internal balance of processes such as sleep, appetite, pain, mood, immune response and inflammation. It has three components: the cannabinoid receptors CB1 and CB2, the endocannabinoids the body makes itself (anandamide and 2-arachidonoylglycerol, or 2-AG), and the enzymes that synthesize and break them down (FAAH and MAGL). Discovered in the 1990s while investigating how cannabis works, the ECS is now regarded as a master regulator of physiological well-being.
Detailed explanation
The ECS operates through "retrograde" signaling: unlike other neurotransmitters, endocannabinoids are produced on demand in the postsynaptic neuron and travel backward to modulate the release of other messengers, acting as a fine brake that prevents both excess and deficiency.
CB1 receptors predominate in the central nervous system (cortex, basal ganglia, hippocampus, cerebellum, periaqueductal grey matter and spinal cord), which explains their role in pain, memory, mood and appetite. CB2 receptors concentrate in immune cells (spleen, macrophages), where they modulate inflammation.
Anandamide is a partial agonist with higher affinity for CB1, while 2-AG activates both receptors equally. Anandamide is broken down by the enzyme FAAH and 2-AG by MAGL. Vincenzo Di Marzo and other researchers have documented how a well-regulated ECS sustains energy, emotional and immune balance. Deficient "endocannabinoid tone" has been associated with chronic pain, anxiety and inflammation. Interventions such as exercise, omega-3 intake, restorative sleep and CBD can modulate this tone.
